How do SNX-BARs form distinct membrane tubules? Do SNX-BARs contribute to a mosaic-like organisation of the tubul ar endosomal network? What molecular complexes couple SNX-BAR tubular sub-domains to cargo sorting? Can SNX-BAR-mediated sorting be reconstituted in vitro? Does PI3,5P2 scaffold the formation of late endosomal sorting complexes? By addressing these aims we will provide fundamental insight into the relationship between the tubular morphology and functional sub-domain architecture of the endosome, and the central dogma of how sorting at the endosomal membrane is co-ordinated with fission, motor-driven transport, uncoating and carrier recognition at the recipient membrane.
